Hi I’ve got a Saab radiator 9-3 manual I’ve noticed an outlet that I would like to use for a header tank connection but think it may have been a vent outlet ? Not sure
I’ve put water in the rad blocked up outlets, water doesn’t come out of this hose tail, but if I remove hose tail water comes out, looking at the hose tail there’s a seal at the back
Can I just cut the seal off and use the hose tail for the header tank
Pretty sure that's the drain plug dude, seals in place and you loosen it a couple turns to let the water out.

Yeah, looks a little different to the Saab auto radiator I have but that’s where I have drain plug is.
Am I right in thinking the feed from the header tank needs to be after the radiator for the water pump to be working properly, at least on the slant engines? I’m sure I read that.

Yes is the simple answer.
To be more precise, the feed from the header tank needs to be plumbed into the cooling system before the water pump,
the best place being the bottom hose. This ensures there is always coolant ready to be circulated by the water pump.
(As this relies on gravity, the best combo is having the header tank as high up as possible allied to a pump situated as low down as practical.)
This is true for all engines, not just slant fours.
